T helper type 2（Th2）細胞
T helper type 2（Th2）細胞は、CD4＋エフェクターT細胞系列に属し、B細胞依存性体液性応答のサポートに加え、腸内寄生虫や細胞外のバクテリアから細胞を保護します。病理学的には、アレルギー性炎症や喘息患者において、Th2細胞活性が見られます。CD4＋エフェクター細胞のTh2細胞への分化は、IL-4, IL-2, IL-7, TSLPなどのサイトカインによって促進されます。Th2細胞は、IL‐4, IL‐5, IL-9, IL-13, IL17E/IL-25を分泌します。

Flow cytometry data showing human peripheral blood naïve CD4+ T cells without (A, C) and with (B, D) a 13 day differentiation using reagents included in the Human Th2 Cell Differentiation Kit. On day 13 of differentiation, the cells were re-stimulated with mitogens and stained with a PerCP-conjugated Mouse Anti-Human IL-17 Monoclonal Antibody (#IC3171C), an APC-conjugated Mouse Anti-Human IFN-gamma Monoclonal Antibody (#IC285A), and a PE-conjugated Mouse Anti-Human IL-4 Monoclonal Antibody (#IC204P). Quadrants were set based on isotype control-stained samples.
分化させたCD4＋細胞のIL-5, IFN-γ, IL-17の分泌量測定
Human peripheral blood naïve CD4+ T cells were differentiated for 13 days using reagents included in the Human Th2 Cell Differentiation Kit. On day 13, cell culture supernatant was collected and cytokine secretion was determined using the Human IL-5 Quantikine ELISA Kit (#D5000B), the Human IFN-gamma Quantikine ELISA Kit (#DIF50、旧バージョン製品), and the Human IL-17 Quantikine ELISA Kit (#D1700).

操作方法概略
- プレートを抗ヒトCD3抗体でコーティングする。
- ヒト血液からPBMC（ヒト末梢血単核細胞）を分離する。
- PBMCからナイーブCD4＋T細胞を単離する。
- 細胞数をカウントする。
- 単離したナイーブCD4＋T細胞を、3～4日ごとに培地を交換しながら、Th2 differentiation mediaで13日間培養する。
- マイトジェンで細胞を再刺激する。
- フローサイトメトリーによりサイトカイン発現を確かめ、Th2細胞へ分化したことを確認する。
