Dipeptidyl Aminopeptidase Inhibitor and Aflatoxin Production Inhibitor Dioctatin A (Hydrochloride)

Dioctatin A was reported as a compound that selectively inhibits DPP2 (dipeptidyl aminopeptidase II) from rat spleen 1,2). And, Dioctatin A has been reported to inhibit aflatoxin production by Aspergillus parasiticus known as aflatoxin-producing fungus, and has potential applications in contamination control against mycotoxin 3). Dioctatin, a synthetic devinyl derivative of dioctatin A, also inhibits aflatoxin production in A. parasiticus, and the target of dioctatin is mitochondrial protease Clp (Chaperonin-linked-protease). The inhibiton mechanism is reported to be that dioctatin-bound ClpP selectively degraded mitochondrial energy-related proteins, including a subunit of respiratory chain complex V 4).

Various subtypes of proline-selective dipeptidyl peptidase (DPP) have been reported, especially DPP4 inhibitors are used for antidiabetic drugs.
Among DPPs, DPP2, DPP4, DPP8, DPP9, and FAP (fibroblast activation protein α) are also called DPP4 Activity and/or Structure Homologue (DASH) proteins 5).
Inhibition or silencing of DPP2, also known as DPP7, has been reported to induce apoptosis of lymphocytes in G0 phase, quiescence stage. Changes in DPP2 activity levels have been observed in many disease states, including autoimmune diseases such as Sjögren's syndrome, rheumatoid arthritis (RA) and lupus erythematosus, Parkinson's disease, neurodegenerative disorders, myopathy, cancer and gastrointestinal disorders 6,7), and there is interest in the relationship between these diseases and DPP2 function.
