R&D Systems offers a Mouse/Rat SOST Quantikine ELISA Kit (#MSST00) suitable for use with cell culture supernates, serum, and plasma. SOST, also known as Sclerostin, is a member of the Cerberus/DAN family of BMP antagonists. It is expressed by osteoclasts in developing bone and inhibits BMP- and Wnt-induced bone formation. Normal bone metabolism is dependent upon the balance between bone deposition by osteoblasts and bone resorption by osteoclasts. These two events are carefully orchestrated by a network of signaling pathways, including the canonical Wnt signaling pathway, which promotes osteoblast development. SOST binds and inhibits the activity of BMP-2, -4, -5, -6, and -7 in vitro . Additionally, SOST has been recently shown to bind to LRP-5/6 and inhibit the canonical Wnt signaling pathway. Inactivating mutations of SOST can cause sclerosteosis and Van Buchem disease, which are bone dysplasia disorders characterized by progressive skeletal overgrowth.
SOST is Secreted by Bone Marrow-derived Mast Cells in vitro. SCF is known for its involvement in the differentiation, survival, and homing of mast cells. Mouse bone marrow cells isolated from non-Swiss albino mice were cultured for 7 days with or without Recombinant Mouse SCF (#455-MC). The Mouse/Rat SOST Quantikine ELISA Kit (#MSST00) was used to determine SOST levels in cell culture supernate. The experiment demonstrates SCF-induced secretion of SOST by differentiated mast cells in culture.
