A disintegrin and metalloproteinase domain-containing protein 12 (ADAM12) is an active metalloprotease that exists in two alternatively spliced forms, a membrane-anchored form and a soluble form lacking the transmembrane and cytoplasmic domains. It is initially synthesized as a pro-peptide that is activated by a furin-like protease in the trans-Golgi apparatus. ADAM12 possesses gelatinase activity and is involved in extracellular matrix degradation, cell adhesion, and intracellular signaling. Increased expression of ADAM12 occurs during pregnancy and in various tumor types, including breast, liver, and bladder cancers.
