Titansphere series enables an effective purification and concentration of phosphorylated peptides.

Background

The study of reversible phosphorylation plays an important role to reveal various mechanisms such as apoptosis, cell cycle and so on.
However, some cases show that the detection of phosphorylated peptide is difficult to detect by mass spectrometry due to its low ionization efficiency and low amount.
To solve these issues, Titansphere series was developed using titania dioxide to absorb phosphorylated peptides specifically.
By using Titansphere series, highly purified and concentrated phosphorylated peptide can be obtained.

Experimental data

Phosphorylated angiotensin II was selectively extracted from mixture of angiotensin II and phosphorylated angiotensin II in only 40 minutes. The recovery rate was 81.6±1.3%.

Titansphere TiO

This material is a porous beads consisting of titania (titanium dioxide) . Posphorylated peptides can be purified and concentrated selectively.

Experimental data

Purified efficiency
Using HeLa cell extracts which contain many impurities, the performance of Titansphere TiO was investigated.
The phosphorylated peptides were detected by about 89.9% in the total identified peptide.
Sample amount : 50 µg
Titania amount : 1 mg/ each

Transphere TiO purified efficiency

Performance test compared to IMAC method
Using arabidopsis thaliana cell extracts, the comparison between Titansphere TiO and IMAC was performed.
The phosphorylated peptides using Titansphere TiO was about 2.6 times higher compared to IMAC on total peak area.
