Static Culture Bag OKUDAKE (OKUDAKE means “Just put it” in Japanese) is a film bag for microbial culture with extremely high gas permeability. It allows sufficient gas exchange even when sealed, enabling static culture of aerobic microorganisms. It eliminates the need for mechanical aeration such as shaking or aerobic stirring, achieving culture without physical stress on microorganisms.

Background

In liquid microbial cultures, gas exchange via mechanical aeration such as shaking or aerobic stirring is routinely performed to supply oxygen, which is essential for survival and yet poorly soluble in water. However, these mechanical operations constantly subject microorganisms to physical stress. To solve this problem, it is necessary to supply oxygen to the culture without mechanical operation, but no effective method has been available so far. To overcome the problem, the “Static Culture Bag OKUDAKE” is made from a film material with extremely high gas permeability.
It enables sufficient oxygen supply to the culture even during static conditions due to its extremely high gas permeability, achieving microbial culture without physical stress.

How to Use

  1. Fill Static Culture Bag OKUDAKE with the culture sample through the opening side.
    * OKUDAKE is delivered non-sterile. If sterilization is required, perform autoclave sterilization or UV sterilization in advance.
  2. Close the opening side of OKUDAKE using a sealing clip or an impulse sealer.
    * If you use an impulse sealer, please verify if it can close OKUDAKE properly, as sealing strength varies by device.
  3. Place the bag flat in an incubator for culture.
    * Avoid applying pressure, as this may cause breakage or leakage.

Application Examples

Static culture of Escherichia coli

Static culture of E. coli using OKUDAKE showed equivalent growth efficiency to conventional shaking culture. Furthermore, when measuring the production yield of the recombinant enzyme (plant-derived peroxidase from Physcomitrium patens), static culture using OKUDAKE showed higher production efficiency than shaking culture.

Biofilm formation in OKUDAKE

Bacillus sp. was cultured under static conditions using OKUDAKE. As a result, they formed a biofilm, whereas they remained dispersed and no biofilm formation was observed in conventional shaking culture.

Static culture of Epicoccum nigrum

Epicoccum nigrum was cultured under static conditions using OKUDAKE. OKUDAKE showed superior growth efficiency compared to shaking culture in a flask. Furthermore, while shaking culture resulted in the formation of spherical pellets, no such pellets were observed in the culture using OKUDAKE.

Physcomitrium patens cultured in OKUDAKE was subjected to microscopic observation under an inverted microscope. Due to the high transparency of OKUDAKE, clear images could be obtained without taking out the plant out of OKUDAKE.

Chlamydomonas culture in OKUDAKE

Chlamydomonas reinhardtii was cultured in OKUDAKE. As a result, the culture using OKUDAKE showed a higher growth rate than that of shaking culture using flasks, and the final biomass yield was also greater. This demonstrates that OKUDAKE is also suitable for algal culture.

Tadpole shrimps sealed in OKUDAKE

Four tadpole shrimps collected from rice fields were sealed in OKUDAKE or a commercially available polyethylene bag with 500 ml of water. As a result, on the second day, all tadpole shrimps sealed in the polyethylene bag had died, while all those sealed in OKUDAKE survived. Furthermore, dissolved oxygen (DO) levels were twice as high in OKUDAKE compared to the polyethylene bag, demonstrating that the high gas permeability of the product supplied sufficient oxygen to the enclosed water.

FAQ

Q1. Is it possible to sterilize before use?

A1. Yes, it can be sterilized with autoclaving or UV. The same sterilization methods for consumables such as plastic tubes and tips can be applied. Although this product is made from high-heat-resistant materials, it tends to soften at around 50–60 °C. During sterilization or drying, if the bag is pressed against an object or bent while heated, it may retain the resulting shape. Handle with care to avoid deformation.

Q2. Can it be used in a low- temperature environment?

A2. Data on the low-temperature resistance is not available, and a lower limit for its usable temperature range has not been established. Since its cold resistance is considered to be lower than that of general plastic films, we do not recommend low-temperature operation with samples inside.
