Application Examples
Static Culture of Escherichia coli
Static culture of E. coli using OKUDAKE demonstrated growth efficiency comparable to conventional shaking culture. Furthermore, when evaluating the production yield of a recombinant enzyme (plant derived peroxidase from Physcomitrium patens), static culture with OKUDAKE achieved higher production efficiency than shaking culture.

Biofilm Formation in OKUDAKE
Bacillus sp. was cultured under static conditions using OKUDAKE. Under these conditions, the cells successfully formed a biofilm. In contrast, when cultured under conventional shaking conditions, the cells remained dispersed and no biofilm formation was observed.

Culture Example of Aspergillus oryzae
Aspergillus oryzae was cultured using this product. The culture grown in OKUDAKE exhibited higher growth efficiency than shaking culture in a flask, and the amount of produced substances was more than twice as high. Furthermore, while shaking culture resulted in the formation of spherical pellets, no pellet formation was observed when cultured using this product, indicating a more uniform and favorable growth environment.

Microscopic Observation of Physcomitrium patens Through OKUDAKE
Physcomitrium patens cultured in OKUDAKE was observed under an inverted microscope. Due to the high transparency of OKUDAKE, clear microscopic images could be obtained without removing the plant from the bag, enabling seamless, contamination free observation.

Chlamydomonas Culture in OKUDAKE
Chlamydomonas reinhardtii was cultured in OKUDAKE. The culture grown in OKUDAKE exhibited a higher growth rate than shaking culture in flasks, and the final biomass yield was also greater. These results demonstrate that OKUDAKE is highly suitable for algal culture, offering an efficient and user friendly alternative to traditional shaking methods.

Tadpole Shrimps Sealed in OKUDAKE
Four tadpole shrimps collected from rice fields were sealed in OKUDAKE or a commercially available polyethylene bag with 500 mL of water. By the second day, all tadpole shrimps sealed in the polyethylene bag had died, whereas all shrimps sealed in OKUDAKE survived.
In addition, dissolved oxygen (DO) levels in OKUDAKE were twice as high as those in the polyethylene bag, demonstrating that the high gas permeability of OKUDAKE supplies sufficient oxygen to the enclosed water and supports organism survival.

How to Use
- Fill the OKUDAKE with your culture sample through the opening.
- Seal the opening using a sealing clip or an impulse sealer.
- Place the bag flat in an incubator for culture.

* OKUDAKE is supplied nonsterile. If sterilization is required, please autoclave or UV sterilize the bag beforehand.
* If using an impulse sealer, ensure that your device can properly seal OKUDAKE, as sealing performance varies by model.
* Avoid applying pressure to the bag, as this may cause damage or leakage.
Why OKUDAKE
Traditional aerobic microbial culture relies heavily on shaking and mechanical agitation to maintain oxygen supply. While effective for aeration, these methods can introduce physical stress that may affect microbial behavior, morphology, and productivity.
OKUDAKE was designed as a new approach to microbial cultivation. Using an ultra gas-permeable film, the bag enables efficient oxygen exchange even during static culture, eliminating the need for mechanical aeration.
The result is a simple, non -shear cultivation environment that supports natural microbial growth, biofilm formation, and recombinant protein production.

FAQ
Q1. Can OKUDAKE be sterilized before use?
A1. Yes. OKUDAKE can be sterilized using standard laboratory methods such as autoclaving or UV sterilization, similar to common plastic laboratory consumables including tubes and pipette tips.
The bag is made from heat-resistant materials; however, the film may soften at approximately 50–60°C. During sterilization or drying, contact with surfaces or bending under heated conditions may cause deformation or shape retention. To maintain bag integrity, avoid applying pressure or folding the bag while it is hot.
Q2. Can OKUDAKE be used at low temperatures?
A2. The lower limit of the recommended operating temperature range for OKUDAKE has not yet been fully evaluated.
Because the film material may have lower cold resistance than conventional plastic films, we do not recommend use under low-temperature conditions with samples inside the bag. To protect your valuable samples from potential bag embrittlement or failure, please avoid low-temperature operations.
Q3. How transparent is the OKUDAKE?
A3. OKUDAKE features a highly transparent film with a haze value below 5% (Haze < 5%), enabling clear visual observation and microscopy through the bag.
Q4. Does OKUDAKE emit autofluorescence when exposed to UV illumination?
A4. No, autofluorescence has not been observed with this product.
The OKUDAKE film exhibits exceptionally high UV transmittance compared to glass and other transparent resins. Because of this property, it does not generate autofluorescence or any other optical interference that would disrupt your microscopic observations or imaging workflows.
Please note that prolonged exposure to UV sterilization conditions may cause slight yellowing of the film. However, this does not affect product performance or usability.
Q5. What are the recommended conditions for sealing the bag with an impulse sealer?
A5. OKUDAKE should be sealed using an impulse sealer capable of temperatures above 230°C with sufficient sealing pressure applied.
Because sealing performance may vary by equipment model, preliminary optimization of sealing conditions is recommended.
Q6. Can OKUDAKE be used to culture organisms other than E. coli?
A6. Yes. OKUDAKE supports a broad range of biological applications beyond E. coli culture.
Successful use has been demonstrated with filamentous fungi, yeast, phytoplankton, mosses, and tadpole shrimp, making the system suitable for diverse cultivation and observation studies.
