Inhibitor of seramidase isolated from Penicillium sp. Ceramidastin

Ceramidastin is a natural bioactive compound which was extracted from a kind of fungi ,Penicillium sp, and inhibits ceramidase in Peudomonas aeruginosa1). It was reported that the ceramidase from ceramidase-producing bacteria causes aggravation of atopic dermatitis. Therefore, it is expected that ceramidastin is useful for various researches such as maintenance of skin barrier function, inflammation control, and lipid metabolism etc., by the inhibition of ceramide degradation1,2).

Influence of bacterial infection between ceramidase activity and regulation of lipid homeostasis

Ceramidase is an important enzyme to degrade ceramide to sphingosine and free fatty acids. Five ceramidases in human are known that acidic ceramidase, neutral ceramidase, alkaline ceramidases 1, 2, and 3. These ceramidases are classified by differentiation of cellular localization and optimum pH.3)
Pseudomonas aeruginosa utilizes skin lipids in human and animal, especially ceramide, during the infection stage. The bacteria induce their ceramidases strongly during biofilm formation. Then, sphingosine and sphingosine-1-phosphate which are metabolites by the enzymes directly involve in additional induction of Pseudomonas aeruginosa ceramidases and inhibition of PPARδ which is a nuclear receptor in skin cells in host animals.4)
In addition, the biofilm formed by Pseudomonas aeruginosa inhibits expression of PPARδ via induction of miR-106b. It has been reported that this low expression level of PPARδ suppresses expression of ABCA12 as lipid-transporter gene, causing imbalance of skin lipid homeostasis and deficiency of skin barrier function.4)

Difference between Ceramidastin and Rubratoxin A

Rubratoxin A harboring a similar structure of ceramidstin is known as a natural bioactive compound extracted from fungus. Both compounds have different functional activities and are noticed that their relationship between their structural difference and activities.

  • Ceramidastin inhibits ceramidase in Pseudomonas aeruginosa but not PP2A in human.4)
  • Rubratoxin A inhibits PP2A in human but not ceramidase in Pseudomonas aeruginosa.1)

Application Note

  • Ceramidastin inhibited Pseudomonas aeruginosa ceramidase (IC50: 6.25 μg/ml) but not ceramidase in human at 200 μg/ml.1)
  • Mouse model of atopic dermatitis showed the increase of mast cells in the dermis by Pseudomonas aeruginosa ceramidase application. Ceramidastin significantly inhibited the increase of mast cells.1).
  • When ceramidastin was administered to mice intravenously, no fatality was observed up to 6.25 mg/kg.1)
  • Ceramidastin did not inhibit human ceramidase at 200 μg/ml.4)
