Foroxymithine was isolated from culture filtrate of Streptomyces as an inhibitor of angiotensin-converting enzyme (ACE, dipeptidyl carboxypeptidase).1)
This compound is a potent chelator of ferric ion (Fe3+) as a siderophore. Utilizing this property, there was an example to protect an administrated compound from the ferric iron in serum.2) A variety of compounds have been reported as siderophores isolated from Streptomyces, it was also reported that the bacteria might utilize foroxymithine in order to obtain ferrous and ferric irons, and take an advantage to survive in their habitat environment.3)

Application Note

  • Among peptidases, foroxymithine selectively inhibited angiotensin-converting enzyme (ACE) derived from bovine lung homoginates (IC50=7 μg/ml).1)
  • Oral administration of foroxymithine (25 mg/kg, 50 mg/kg) to spontaneously hypertensive rats (SHR) resulted in a reduction of systolic blood pressure. The maximum value was decreased in 1-3 hours after dosing.1)
  • Erbstatin is a tyrosine kinase inhibitor which is degraded in the presence of ferric irons (Fe3+) in serum. When used in assays together with erbstatin, foroxymithine suppressed the iron-induced degradation of the tyrosine kinase inhibitor.2)
