Bioactive compound harboring anti-tumor and anti-angiogenesis activities NM-3 [CAS: 181427-78-1]

NM-3 harboring isocoumarin backbone is a synthesized derivative of cytogenin (#16331), which is redesigned to have biologically stable structure. A variety of the derivatives were examined and concluded that NM-3 had the most superior pharmacological activity than other derivatives.1, 3) In addition, it was reported that oral administration of NM-3 to mice showed dose-dependently suppression of angiogenesis.3)

Application Note
- Oral administration of NM-3 (0.310 mg/kg/day) to mice dose-dependently suppressed angiogenesis induced by S-180 tumor cells.3)
- The combined treatment with NM-3 and chemotherapeutic agents (including 5-FU, docetaxel and ionizing radiation) significantly reduced mean tumor volume (including HT29, MKN28, MCF-7, LLC and A549) compared to either treatment alone, with no effects on body weight changes.4~6)
NM-3, biologically more stable than Cytogenin
Cytogenin was first isolated from fermentation broth of Streptoverticillium eurocidicum as an immunomodulative antitumor substance 7), and had an anti-inflammatory activity against collagen-induced arthritic model in mice.2, 3)
However, it was reported that administrated cytogenin to mice was converted to glucuronate or sulfate conjugated forms with low activity easily, or another compound (MC-1) which was substituted to carbonate at the position 3 carbon in cytogenin, during metabolism. This phenomenon caused a decrease in the activity of cytogenin in animal testing. In order to prevent generation of the conjugated forms, O-acyl, and O-alkyl derivatives, and other analogues harboring carbonate at the position 3 sidechain. NM-3 is the derivative showing the best pharmacological activity among them.
