Phepropeptin is a bioactive cyclic peptide and isolated from cultured broth of Streptomyces by screening of proteasome inhibitor activity. Four types of Phepropeptin having different amino acid residues at the side chain were discovered.
The cyclic peptides have selective inhibitory activity to proteasomal chymotrypsin-like activity. In contrast, they showed weak inhibition to trypsin-like activity and peptidyl glutamyl peptide hydrolyzing activity. They did not inhibit alpha-chymotrypsin activity. 1) Then, it was also reported that the cyclic peptides have high cell permeability. 2)

Application Note
- It was reported that the cyclic peptides showed inhibitory activity to proteasomal chymotrypsin-like activity from mouse liver, but not inhibit trypsin-like activity up to 100 ug/ml. In addition, they had weak inhibitory activity to peptidyl glutamyl peptide hydrolyzing activity (IC50 value 21.0 ug/ml). It showed no inhibition toward alpha-chymotrypsin from bovine pancreas at 100 ug/ml. 1)
