Benastatin A is isolated from the culture broth of Streptomyces sp. MI384-DF12 as an inhibitor of glutathione S-transferase (GST).1,2) The bioactive compound has been reported to inhibit GST class Phi (GSTF), which is involved in detoxication and stress response.3) It has antibacterial activity against gram-positive bacteria including methicillin-resistant Staphylococcus aureus (MRSA).1)

Application Note

  • The inhibition of benastatin A against GST is competitive with 3,4-dichloronitrobenzene (Ki: 5.0 × 10-6 M), and noncompetitive with glutathione (Ki: 3.5×10-6 M).1)
  • Benastatin A has activity against gram-positive bacteria including methicillin-resistant Staphylococcus aureus (MRSA).1)
  • The compound exhibited an inhibitory activity to Micrococcus luteus (MIC=31.3 μM).4)
  • In rat basophilic leukemia-derived RBL-2H3 cells, an evaluation model for inhibition of immediate-type allergic hypersensitivity reactions, the compound inhibited IgE-stimulated β-hexosaminidase release (IC50=79 μM) and suppressed mast cell degranulation.4)
  • The compound inhibited OsGSTF5 activity which is rice-derived (IC50=1.36 μM).3)
