Cell Function & Temperature

Temperature is physical quantities that controls all chemical reactions in cells. Chemical reactions involving exothermic or endothermic reactions at a specific site in cells to carry out cell-function induce local temperature-change. Therefore, the intracellular temperature distribution reflects the thermodynamics and function of intracellular molecules. From the medical perspective, it has been reported that pathological cells such as cancer cells have enhanced thermogenesis. Therefore, understanding the intracellular temperature distribution is expected to contribute to a better understanding of cellular functions and to the development of new diagnostic and therapeutic methods.

Features

  • Temperature detection limit is 0.18-0.5℃, which enables detection of even the smallest temperature differences.
  • Spatial resolution is 200 nm and can detect temperature differences in intracellular organelles.
  • Temperature measurements with Diffusive Thermoprobe are negligibly affected by intracellular ionic strength, pH, protein content and viscosity.
  • Since measurement is performed by fluorescence lifetime, temperature can be accurately measured without being affected by local concentration of fluorescent probe, excitation light intensity, fading, excitation wavelength, etc.
  • Solubility: Soluble in water

Structure & Principles

Structure & Principles for Diffusive Thermoprobe<sup>™</sup>

Diffusive Thermoprobe is composed of thermosensitive unit (NNPAM), hydrophilic unit (SPA) and fluorescent unit (DBD-AA) . When a probe solution is at low temperature, fluorescent is weak due to the presence of water molecules in the structure. However, at high temperature, the hydrophobic interaction of thermosensitive unit causes the probe to become small and rounded, so that water molecules are excluded from the probe and strong fluorescence is emitted. In addition, the incorporation of hydrophilic unit prevents aggregation of fluorescent temperature sensors within a cell, enabling temperature distribution measurements that require high spatial resolution.

Outline of Protocol

  1. Reconstitute 100 μg powder at 10 μl of 80 mM KCl, 10 mM K2HPO4, 4 mM NaCl.
  2. Leave it at +4℃ at least 8 hours.
  3. Take 1 μl solution from vial, and fill it up to glass capillary needle for microinjection.
  4. Microinject it into cytoplasm with a glass capillary needle below 30℃.
  5. Leave cells for 30min.
  6. Analyze intracellular temperature by fluorescence lifetime imaging microscopy.
