Cell Function & Temperature

Temperature is physical quantities that controls all chemical reactions in cells. Chemical reactions involving exothermic or endothermic reactions at a specific site in cells to carry out cell-function induce local temperature-change. Therefore, the intracellular temperature distribution reflects the thermodynamics and function of intracellular molecules. From the medical perspective, it has been reported that pathological cells such as cancer cells have enhanced thermogenesis. Therefore, understanding the intracellular temperature distribution is expected to contribute to a better understanding of cellular functions and to the development of new diagnostic and therapeutic methods.

Strong Point

  • Particulate Thermoprobe is a nanogel-type fluorescent temperature probe that aggregates in a temperature-dependent manner after being introduced into cells, and its fluorescence intensity depends on temperature. By measuring the fluorescence intensity of Particulate Thermoprobe in a single cell, the average temperature of the cell can be calculated.
  • Temperature detection limit is 0.5℃ or less, allowing detection of even the slightest temperature difference.
  • Particulate Thermoprobe is not affected by cellular environmental conditions such as pH, ionic strength, protein content, and viscosity etc.

Structure & Principles

Structure & Principles for Particulate Thermoprobe<sup>™</sup>

Particulate Thermoprobe is composed of thermo-responsive unit (polyNIPAM), water-sensitive fluorescent unit (DBThD-AA) and crosslinking unit (MBAM). When a probe solution is at low temperature, water molecules are absorbed into Particulate Thermoprobe and the fluorescence of DBThD-AA inside is quenched by the surrounding water molecules. As temperature rises, Particulate Thermoprobe contracts and emits fluorescence as the water molecules are released.

Outline of Protocol

  1. Reconstitute 100 μg powder at 10 μl of 80 mM KCl, 10 mM K2HPO4, 4 mM NaCl.
  2. Leave it at +4℃ at least 8 hours.
  3. Take 1 μl solution from vial, and fill it up to glass capillary needle for microinjection.
  4. Microinject it into cytoplasm with a glass capillary needle below 30℃.
  5. Leave cells for 30min.
  6. Analyze intracellular temperature by fluorescence microscopy.
