ET-1（Endothelin-1）とは
ET-1（Endothelin-1）とは
エンドセリン-1（ET-1、Endothelin-1）は、21アミノ酸残基のペプチドより成り、強力な血管収縮剤としての作用で知られる多面発現分子です1。ET-1は、エンドセリン-2（ET-2）およびエンドセリン-3（ET-3）を含む異なる遺伝子によってコードされる3つのタンパク質ファミリーの1つです2,3。ET-2とET-3は、ET-1とそれぞれアミノ酸2個、およびアミノ酸6個が異なります1,2。これらのエンドセリンは、すべてC末端に2つの必須のジスルフィド架橋と6つの保存されたアミノ酸残基を含んでいます。
ET-1（Endothelin-1）の生合成
ヒトET-1は、初めに212アミノ酸のプレプロポリペプチドとして合成されます2,4。シグナルペプチダーゼにより、タンパク質分解的に切断されてプロET-1を生成し、さらにフューリン様プロテアーゼによって処理されてBig ET-1が生成されます。次に、Big ET-1は、膜結合型メタロプロテアーゼであるエンドセリン変換酵素（ECE-1）によって切断され、強力な成熟型ET-1 を生成します5,6。血管内皮は、ET-1の豊富な供給源です 3、7。また、白血球、平滑筋細胞、メサンギウム細胞、心筋細胞、星状細胞によっても発現する可能性があります8,9。
ET-1は、機械的刺激、様々なホルモン、炎症誘発性サイトカインなど、多くの要因によって内皮細胞に誘導されます。一酸化窒素（NO）、環状ヌクレオチド、プロスタサイクリン、心房性ナトリウム利尿ペプチド（ANP）によって産生が阻害されます10,11。
ET-1（Endothelin-1）の機能と疾病への関与
ET-1は、心臓の収縮と心筋細胞の成長を刺激し、血管作用物質の放出を調節し、平滑筋細胞の有糸分裂誘発を刺激します。またET-1は、好中球の接着と遊走を促進し、炎症性サイトカインの産生を刺激することにより炎症反応を制御する可能性があります。さらに、腫瘍細胞の増殖と移動を調節し、血管新生促進因子として作用するがんの進行にも関与しています13。ET-1は、敗血症性ショック、アテローム性動脈硬化症、心不全、腎不全、肺高血圧症、くも膜下出血に関連する脳血管状態など、他の病状において役割を果たすとも考えられています9。
