Product Summary

The Quantikine Human BDNF Immunoassay is a 3.5 hour solid phase ELISA designed to measure human BDNF in cell culture supernates, serum, and plasma. It contains recombinant human BDNF expressed in Sf 21 cells and antibodies raised against the recombinant factor. This immunoassay has been shown to quantitate the recombinant BDNF accurately. Results obtained measuring natural human BDNF showed dose-response curves that were parallel to the standard curves obtained using the recombinant Quantikine kit standards. These results indicate that this kit can be used to determine relative mass values for natural human BDNF.

Linearity

To assess the linearity of the assay, five samples containing or spiked with high concentrations of BDNF in various matrices were diluted with the appropriate Calibrator Diluent to produce samples with values within the dynamic range of the assay.

BDNF
Brain-derived neurotrophic factor (BDNF) is a member of the NGF family of neurotrophic factors (also named neurotrophins) thatare required for the differentiation and survival of specific neuronal subpopulations in both the central as well as the peripheralnervous system. The neurotrophin family is comprised of at least four proteins including NGF, BDNF, NT-3, and NT-4/5. Thesesecreted cytokines are synthesized as prepropeptides that are proteolytically processed to generate the mature proteins. Allneurotrophins have six conserved cysteine residues that are involved in the formation of three disulfide bonds and all shareapproximately 55% sequence identity at the amino acid level. Similarly to NGF, bioactive BDNF is predicted to be a non-covalentlylinked homodimer.BDNF cDNA encodes a 247 amino acid residue precursor protein with a signal peptide and a proprotein that are cleaved to yieldthe 119 amino acid residue mature BDNF. The amino acid sequence of mature BDNF is identical in all mammals examined. Highlevels of expression of BDNF have been detected in the hippocampus, cerebellum, fetal eye and placenta. In addition, low levels ofBDNF expression are found in the pituitary gland, spinal cord, heart, lung and skeletal muscle. BDNF binds with high affinityand specifically activates the TrkB tyrosine kinase receptor.
