Metallo Assay Zinc Assay Kit LS (5-Br-PAPS method ) can measure Zinc ion (Zn2+) in the biological samples such as Plasma, Serum, Urine, and wide range of samples by microplate reader.
Background
Zinc is a cofactor of more than 200 kinds of metalloenzymes, and is also a trace element concerned in synthetic of ribonucleic acid or protein. It is widely known that acute zinc deficiency during the growth stage of the mammalian, results in a severe impairment of the skin or hair, and may lead to arrested development. Zinc is essential to reproduction of cell and its relevant supply is necessary for healthy growth.
Features
- Using microplate reader for assay, no need to use Atomic Absorption Spectrometry or ICP-OES Optical Emission Spectrometry, which have been mainly used so far.
- Good correlation with the result of Atomic Absorption Spectrometry and ICP-OES.
- High throughput measurement by using microplate reader.
- All animal species sample can be measured.
- Assay time : 10 minutes (except sample preparation time)
- 1 point calibration – do not need multiple point measurements to make standard curve (r2=0.9999).
- Samples : Serum, Plasma, Urine, Saliva, Cell lysate, Tissue extract, Hair extract, Water, etc
Assay Principle
This kit measures zinc concentration by color change caused by chelate complex formulation of 5-Br-PAPS and Zinc. Zinc combined to transportation protein is dissociated by denaturating agent, then formulates Zinc-5-Br-PAPS chelate complex. Measuring this complex by 560 nm wavelength and obtain zinc concentration.
Handling procedure
| (1)Apply DW, Standard and Sample (12 uL) to 96 well plate. |  |
| (2)Add Working reagent (R-A Buffer + R-R Chelate color ; 230 uL). |  |
| (3)Incubate at room temperature for 5 minutes. |  |
| (4)Read the absorbance at 560 nm (main) and 700 nm (sub) . |  |
| (5)Calculate concentration.
