AllView PAGE Buffer (20×) is an innovative running buffer designed for protein SDS-PAGE. By simply replacing your conventional running buffer with this product, you can achieve wide-range protein separation across a broad molecular weight scale using standard, self-cast Laemmli gels—simulating the performance of a gradient gel.
Note: It is optimized for self-cast Laemmli gels and may not provide sufficient separation when used with precast gels.

Features

No Gradient Gel Required

This buffer eliminates the need to purchase precast gradient gels. Using a standard self-cast 6% polyacrylamide gel with AllView PAGE Buffer delivers a fractionation range equivalent to that of a gradient gel.

Comparison of Gradient Gel and AllView PAGE Buffer

Self-cast 6% polyacrylamide gel (5% C)*1 and DynaMarker Protein MultiColor Stable II (#DM660)*3 are utilized.

*1 If using precast gels, the required gel concentration may differ or protein separation may be insufficient.
*2 Conventional running buffer: Tris-Glycine SDS buffer
*3 Apparent molecular weights may vary depending on the lot and running conditions. For details, see the datasheet attached to the product.

High Blotting Efficiency

Because it utilizes a low-concentration self-cast gel (6%), it offers higher western blot transfer efficiency compared to standard gradient gels. This ensures smoother and more complete protein transfer to PVDF membranes.

Transfer method: Semi-dry blotting using a continuous buffer system (Towbin buffer containing 20% MeOH and SDS) and a PVDF membrane (0.2 μm)
Conditions: 2 mA/cm2, 60 min

Wide-Range Separation on Any Gel Size

  • Wide Fractionation: Successfully resolves proteins ranging from 8 kDa to 230 kDa on a single gel concentration.
  • Midi-Gel Compatible: Works flawlessly with both mini-gels and large midi-gels (e.g., 140W × 140H mm)

High-Speed Electrophoresis

  • Time-Saving: Cuts total run time down to approximately 1/3 of traditional Tris-Glycine SDS buffers.
  • Run Times (at 250V constant):
    Mini-gels: ~15 minutes.
    Midi-gels: ~50 minutes.

Technical Information: Quick Gel Preparation Recipe

By adding glycerol to the separating gel, you can cast both the separating and stacking layers simultaneously without waiting for the first layer to solidify.

① Separating gel solution (6% concentration, for 2 mini-gels)

Glycerol1.5 mL
Ultrapure Water6.55 mL
1.5M Tris-HCl (pH 8.8)3.75 mL
30% Acrylamide / Bis solution3.0 mL
10% SDS0.15 mL
TEMED3.75 μL
10% APS0.05 mL

② Stacking gel solution (3% concentration, for 2 mini-gels)

Ultrapure Water3.9 mL
0.5M Tris-HCl(pH6.8)1.5 mL
30% Acrylamide / Bis solution0.6 mL
10% SDS0.06 mL
TEMED3 μL
10% APS0.06 mL

③ Gel Polymerization

  1. Pour the prepared separating gel solution (with glycerol) into the glass plate cassette.
  2. Immediately and gently layer the stacking gel solution directly on top.
  3. Allow it to sit at room temperature for approximately 1 hour to polymerize.
    Tip: Setting it inside a 37°C incubator reduces polymerization time to about 30 minutes.
