This size-exclusion chromatography column purifies exosomes from samples such as serum, plasma, and cell culture supernatant in high purity by gravity fall.
Because it elutes in PBS without ultracentrifugation, there is no structural damage, and the purified exosomes can be used in a wide range of applications.
EVSecond L70 significantly reduces the mixing time and can be operated at room temperature.

Procedure
(A) Preparation of Column (To be executed in RT condition)
Mix filler well by inverting column until filler is mixed evenly and completely.
* Mix the column several minutes gently in RT is recommended.
* Filler clumps may cause incorrect fraction. Check no clumps are found before use.
(B) Purification (To be executed in RT condition)
- Remove two caps (top and bottom) and discard storage buffer.
- Block the filler by 700 μL of FBS (filtrated by 0.22 μm)
- Wash 3 times by 1500 μL of PBS
- Apply 50 to 1500 μL of sample. Flow all sample completely.
- Flow proper amount of PBS
- Collect exosomes fraction
Attention
* Initially, elution position of exosome and serum proteins should be checked by anti-CD9-anti-CD63 sandwich ELISA (for exosome) or anti-CD9 western blot (for serum proteins).
Special Rack: GL-EXO Fraction Rack
(1)Column hanger
Maxium 6 columns can be installed.
(2)Collection tube racks
Maxium 8 collection tubes(1.5 mL or 2 mL) can be set.
The collectcion tube can be switched by pulling the lever.
(3)Base plate
Maximum 6 collection tube racks can be set.
