Laminin is a basic component of basement membrane, composed of alpha, beta and gamma subunits. Laminin is also known as a superior cell culture substrate, and its bioactive sequence interacts with integrin, syndecan, etc. It has been thought that specific laminin receptor's function analysis is difficult as various laminin receptors interact with laminin in parallel during cell adhesion.

AG73, A208, C16 and A3G756 are synthetic peptides which have bioactive sequence of laminin. These peptides can be used as a cell culture substrate as they bind to each receptor specifically. Cell function behavior, induced by its specific receptors and downstream signaling can be analyzed by using these peptides.

Recombinant laminin alpha1 chain (LG4) or AG73 (specific region of LG4) is used as a coating material. Recombinant laminin alpha1 chain (LG4) contains Integrin α2β1 or syndecan binding site, but only AG73 binds to syndecan. Morphology is very different between two coating materials.


Concentration-dependent cell adhesion by each peptide: Each peptide is coated on 96 well culture plate and blocked by BSA. Human Dermal Fibrobrast is cultivated (20,000 cells/well), incubated two hours and fixed by 4% paraformaldehyde. Cells are stained by crystal violet and counted its numbers.
