What is Thiol Group

Thiol groups exist in the body as proteins such as albumin, glutathione, and free thiol groups. Thiol has a strong reducing ability and rapidly reacts with reactive-oxygen-species (ROS) to eliminate ROS and form disulfide. Protein thiol groups are more susceptible to oxidative modification than other amino acid side chains, and the function of some proteins and enzymes is controlled by the redox state of thiol groups.
Serum thiol concentration is an indicator of the redox environment in the body, and is also attracting attention as a marker of oxidative stress. It has been reported that serum thiol concentration decreases in various diseases such as cancer and liver disease, suggesting that serum thiol concentration may be an indicator of diseases and aging.

Ellman's Method

Ellman's reagent (DTNB: 5,5'-Dithiobis(2-nitrobenzoic acid)) reacts with thiol groups in blood samples, and yields TNB2- (5-Mercapto-2-nitrobenzoic acid), showing yellow color. The intensity of the color is proportional to the thiol groups in blood samples. Thiol concentration can be calculated by measuring absorbance of TNB^2- at wavelength 412 nm (380 nm to 440 nm).
