A mouse monoclonal antibody that specifically recognizes asymmetric dimethylated arginine (R216, R218) in TLS/FUS (Translocated in LipoSarcoma / Fused in Sarcoma). It does not cross unmethylated TLS/FUS or symmetrically dimethylated TLS/FUS. TLS/FUS is said to be one of the causative genes of amyotrophic lateral sclerosis (ALS).

  • Lane1: Recombinant GST-TLS/FUS methylated using PRMT1 in the presence of SAM (20 mM)
  • Lane2: Cell extract from CHO cells immunoprecipitated with rabbit polyclonal anti-TLS/FUS antibody (Bethyl Laboratories, #A300-302A).

About Asymmetric Dimethylarginine of TLS/FUS

Arginine methylation is one of posttranslational modifications, catalyzed by Protein Arginine Methyltransferases : PRMT. Arginine is known to be methylated once (monomethylated arginine) or twice (dimethylated arginine). Dimethylation can be further categorized asymmetric (me2s) or asymmetric (me2a).

TLS/FUS (Translocated in LipoSarcoma/Fused in Sarcoma) is known as one of causative genes for familial amyotrophic lateral sclerosis (ALS). Especially, it is reported that four arginine residues in TLS/FUS (R216, R218, R242, and R394) are constitutively dimethylated by protein arginine methyltransferases.
It is also reported that TLS/FUS binds to lncRNAs transcripted from cyclin D1 promotor. However, the function of methylated TLS/FUS is still unclear. This antibody recognizes asymmetric dimethylarginine (R216, R218) of TLS/FUS. It does not cross-react against symmetric dimethylarginine of TLS/FUS, and non -methylated TLS /FUS.

Features

  • This antibody can detect asymmetric dimethylarginine (R216, R218) of TLS/FUS specifically by westernblot in sample which is immunoprecipitated by Bethyl laboratories' anti-FUS antibody (A300-302A)*.
  • Immunoprecipitation of samples with this antibody allows the function of methylation of TLS/FUS to be analyzed by RNA-binding protein immunoprecipitation.

*Since it is difficult to specifically detect dimethylated TLS/FUS in cell lysate samples by Western blotting, it is recommended to perform immunoprecipitation with anti-TLS/FUS polyclonal antibody beforehand.
