This product is a clodronate-incapsulated liposome reagent. Clodronate itself is known to be difficult to pass through a cell membrane and to be incorporated by macrophage through phagocytosis. Clodronate liposome has efficient cell permeability, enhances macrophage phagocytosis, and can be used in macrophage depletion experiments.

What is clodronate?

Clodronate is a compound that depletes osteoclasts and inhibits bone absorption, and can be used in osteoporosis research. This compound is known to inhibit ATP metabolism as ATP analogue and to deplete not only osteoclasts but also macrophages.
In recent years, it has been reported that macrophages are associated with various diseases such as cancers, diabetes, and arteriosclerosis. To study relationship between such diseases and macrophages, it is necessary to temporarily deplete macrophages and observe changes of the state. Therefore, clodronate has recently been gaining attention as a useful tool to study these diseases.


Features

  1. This product has lipid composition to be easily incorporated by macrophages and induces depletion of the cells temporally.
  2. This product is sterilized and tested that it is stable for two years from a production date checked by stability testing at 4 degrees.
  3. Two types of the products are available that one is a large particle type with 300nm diameter for targeting reticuloendothelial macrophages in liver, spleen etc., and the other is a small particle type with 100 nm diameter called SMALL for targeting tumor-associated macrophages.
  4. Internal data for administration route, dosage, and duration of effect of the products are available. In addition, many product citations are also available. (See Application note part below.)
  5. Internally used cell strains : Balb/c, C57BL/6J, C57BL/6, DBA/2, Balb/cA Jcl-nu/nu, and NOD/SCID/IL-2Rg null (NOG)
  6. Internally used model mice: Normal mice, gallbladder cancer model mice, and inflammation model mice

Application note

Induction of macrophage depletion in the liver and the spleen of normal mice

Normal mice were intraperitoneally administrated 200ul of the product. The liver and spleen were removed 24 hours later and expression levels of macrophage markers F4/80 and CD68 were checked by real-time PCR.

Immunostaining of macrophages in the liver and the spleen of normal mice

Normal mice were intraperitoneally administrated 200ul of the product. The liver and spleen were removed 24 hours later, and localization of macrophages was observed by tissue immunostaining.

Macrophage depletion effect in liver and spleen of gastric cancer model mice (1)

* The data were obtained by using Clodronate liposome (SMALL).
200ul of the product was administrated to tail vein of gastric cancer model mice. Tissue immunostaining of F4/80 was performed on the extracted liver and the spleen on day 3 or day 5 from the administration.

* The data were obtained by using Clodronate liposome (SMALL).
The tumor tissues of gastric cancer model mouse were extracted on the fifth day from tail vein administration. HE staining and immunostaining of F4/80 were performed on the extracted tissues.
