About Tyrosinase
Tyrosinase (EC1.14.18.1) is the primary metabolic enzyme for melanin synthesis existing in a wide variety of organisms including bacteria, plants, insects, and animals. Melanin is synthesized in pigment cells (melanocytes) in skin and hair apparatus mainly in vertebrates and protects cells from ultra-violet (UV) radiation. On the other hand, excessive exposure of UV rays or physical stress induces excessive melanin production, which causes skin blemishes by melanin pigmentation. Long term preservation or damages to the plant body in edible plants cause tyrosinases to promote melanin production and induce discoloration. It was reported that some substances which inhibit tyrosinase activity give whitening effect and inhibition of browning. In contrast, other substances which promote the enzymatic activity suppress hair graying.
In melanin synthesis, tyrosinase catalyze two reactions mainly. One is monophenolase activity to catalyze oxidation of monophenol such as L-tyrosine to quinones such as L-Dopaquinone, the other is diphenolase activity to catalyze oxidation of diphenols such as L-DOPA etc. to quinones. Although sensitivities toward substances of tyrosinases are different between monophenolase and diphenolase activities, tyrosinase activities were measured by generally using L-DOPA as a substrate which can prepare easily in many cases, it was difficult to measure the enzymatic activities by using hardly soluble L-tyrosine as a substrate.
These kits can validate monophenolase or diphenolase activities from human, mouse, or mushroom, stably by using L-tyrosine or L-DOPA as a substrate.

- The kits can be used for screening of inhibitors or enhancers of the tyrosinase which is the primary enzyme in the melanin synthetic pathway.
- The kits can be selected to measure activity of tyrosinase from human, mouse, or mushroom. They are the first assay kits to measure mammalian tyrosinase activity in the reagent market.
- The kit can select L-tyrosine or L-DOPA as a substrate and measure two individual enzymatic activities.
Product Lineup
Mammalian tyrosinase and mushroom tyrosinase have different structure and localization. In addition, mouse tyrosinase and human tyrosinase have different sensitivity to substances because of their different enzymatic functions.
It is recommended to use human tyrosinase for screening of whitening substances and mushroom tyrosinase for screening for anti-browning substances.

Protocol Overview
- Add 20μL of samples to each well on 96 well microplate.
- Add 20μL of Enzyme solution to each well and incubate at 25-30℃ for 10 minutes.
- Add 60μL of Substrate solution to each well and incubate at 25-30℃ for 30 minutes.
- Measure the absorbance at wavelength 495nm in each well.
Application Note
Verification of inhibitory activity of Kojic acid by individual kits for different enzyme activities and substrates
Chromogenic reaction was performed for control (untreated), 0.0032, 0.016, 0.08, 0.4, 2, and 10 mM of kojic acid, respectively.
Measurement of 50% Inhibitory Concentration (IC50: μM) to representative substances for whitening and anti-browning activities.
