Mazethramycin B (mazethramycin 11-methyl ether) harboring antitumor and antibacterial activities was isolated from culture filtrate of Streptomyces thioluteus in Institute of Microbial Chemistry (BIKAKEN) in 1980.¹⁾ Mazethramycin species were reported that there were two compounds as mazethramycin A and B. The institute developed mazethramycin B having more stable structure than mazethramycin A, as a research reagent.

Structure fo Pyrrolobenzodiazepine (PBD)

Mazethramycin species are natural compounds which have a charaseristic pyrrolobenzodiazepine (PBD) structure harboring a tricyclic structure composed of seven-membered ring. Many of these natural compounds harboring the PBD were discovered in Streptomyces. Some compounds harboring PBD structure exhibited antitumor activity by sequence-selective DNA alkylation.²⁾ In addition, development of antibody-drug conjugation (ADC) has been progressing to use PBD structure as cytotoxic poison (payload).³⁾

Application Note

Mazethramycin B administration prolonged the survival period of mice inoculated with leukemia L-1210 (T/C: 240% at 31.3 μg/kg/day).¹⁾
This compound exhibited antibacterial activity against gram-positive and gram-negative bacteria.¹⁾
Intravenous injection of the compound in mice showed low toxicity (LD₅₀: 0.8 mg/kg).¹⁾
