Clazamycin A which has basic structure of pyrrolizidine was isolated from culture filtrates of Streptomyces. It was reported that this compound had a weak antibacterial activity and extended survival duration of mice inoculated leukemia L-1210 cells by administration of it everyday for 10 days.1) In addition, this compound inhibited DNA synthesis, membrane transport of some specific amino acids, and other nutrients in Escherichia coli.2)

Application Note

  • Intravenously administration of clazamycin A (12.5~100ug) extended 30% of survival duration of mice which were inoculated leukemia L-1210 cells.1)
  • Clazamycin A inhibited the membrane transport of N-acetylglucosamine and leucine.2)
  • Clazamycin A inhibited DNA synthesis at the polymerization step in E. coli.2)

Equilibration between clazaymcin A and B

  • Clazamycin A can be changed to its isomer clazamycin B in aqueous solution in a pH dependent manner and form the equilibrium mixture of them.1, 3)
  • Although this product contains less than 5% of clazamycin B, it is possible to prevent decomposition and isomerization to clazamycin B at pH 3 or lower.
  • In order to avoid the decomposition and isomerization, it is recommended to resolve into the acidic solution, and store at 4 degree or lower. They should be used within a couple of days.
