Actinase E is a high-purity enzyme that removes contaminating proteins by strong proteolytic action.

What is Actinase E

Actinase E is a proteolytic enzyme product derived from the culture filtrate of the actinomycete Streptomyces griseus. It was developed in Japan. Today, it is widely used in research around the world to remove protein/peptide components from samples such as sugars and nucleic acids.

Features

  • Low amino acid specificity and cleaves peptide bonds almost indiscriminately
  • High enzyme activity in the neutral range (pH7-9)
  • Unlike acid hydrolysis, it does not degrade tryptophan/methionine, allowing protein degradation even under mild reaction conditions.
  • The enzyme itself is inactivated by increasing temperature, however it can be reacted at high temperatures (50-55°C) in the presence of substrate.
  • Strong proteolytic activity (1,000 Tyrosine PU*/mg), and the degree of degradation reaches 75% to 90%.
    PU: Enzyme activity that releases 1 μg of tyrosine per minute using casein as substrate

  • Heat, acid, alkali, serum, potato and legume extracts, and the presence of rich salts cause a decrease in activity. There is no problem using normal saline (0.9% NaCl).

    Because the enzyme is calcium-dependent, the enzyme is rapidly deactivated when calcium ions are lost by treatment of the reaction mixture with EDTA, dialysis, ammonium sulfate, or ion exchange resin.
