Electrophoresis using 12.5% acrylamide - 7.5 M urea denaturing gel

Each band of RNA ladder marker is labeled with DIG (digoxigenin) together with blue dye.
In addition to confirming transfer efficiency to a membrane, it can also be used as a molecular weight marker in Northern hybridization using DIG-modified probes.

Background

To confirm the molecular weight during Northern hybridization under non-RI conditions, the gel portion containing the marker lane was cut off and stained with EtBr, or after transfer to a membrane, stained with methylene blue or other dyes 1. Since this product is DIG labeled, it can be detected using DIG antibody. In addition, since it is modified with visible light dye, it can be used for monitoring electrophoresis and checking transfer efficiency to membrane.

Festures

  • It is very useful for Northern hybridization of small RNA such as miRNA.
  • It is ideal for electrophoresis and monitoring transfer efficiency to membranes (Fig. 1).
  • DIG is introduced into the colored nucleic acid that constitutes the marker. It can be detected by using alkaline phosphatase-modified DIG antibody and chemiluminescent substrate (Fig. 2).
  • High concordance rate was achieved with Dyna Marker Small RNA II (#DM192), which is a molecular weight marker for electrophoresis of small RNA.
  • It can be applied directly without heat denaturation.
