We introduce the p53 antibody, which is the most important cancer-related gene detected in more than half of human cancers, and related antibody products involved in repairing DNA damage and inducing apoptosis.
Background
p53 and Ser46 phosphorylated p53
The p53 protein is the product of the tumor suppressor gene p53. It is a 393 amino acid protein with a molecular weight of 53 kDa. The p53 protein consists of a transcriptional activation domain and MM2 binding domain in the N-terminal region, a core domain with DNA-binding ability in the middle, and a tetramer-forming domain in the C-terminal region.
p53 is the most important cancer-related gene, with mutations detected in more than half of human cancers. The p53 protein is inactive in the absence of stress, but is activated by various stresses, especially DNA damage, and is involved in cell cycle arrest, repair of DNA damage, and induction of apoptosis. The p53 protein is activated by phosphorylation of multiple phosphorylation sites in the N-terminal and C-terminal regions, and by acetylation of lysine in the C-terminal region.
Among the phosphorylation sites, Ser46 is a site that is phosphorylated when DNA damage is too large to be repaired, and is reported to activate transcription of p53AIP1, a p53 target gene, and induce apoptosis. It has also been suggested that HIPK2, known as a transcription-coupled regulator, may be involved in phosphorylation of Ser46.
E2F1 and MdmX
In response to DNA damage, E2F1 is phosphorylated at serine 364, stabilized, and translocated to the nucleus where it is activated as a transcription factor. E2F1 activates the transcription of p73, a homolog of p53, and induces apoptosis.MdmX binds to the transcriptional activation domains of p53 and p73 and inhibits cell-cycle arrest and apoptosis that depend on them. Phosphorylation of MdmX at Ser367 in response to DNA damage increases its binding to 14-3-3 and translocates to the nucleus. There, it is degraded by Mdm2, which activates p53.
