Extrap appearance

The Extrap Soil DNA Kit Plus extracts and purifies microbial DNA from a wide range of environmental samples, including soil and activated sludge. It is suitable for applications such as community structure analysis and real-time quantitative PCR of microorganisms present in environmental samples. Using this kit, DNA can be extracted with high yield even from Andosol*1, which has high DNA adsorption properties.
In addition, the magnetic bead format enables automated multi-sample processing.

*1 Andosol (Andisol/Volcanic ash soil): Soil with high DNA adsorption. Andosols are rare worldwide, accounting for less than 1% of the total land area.

Features

  • Microbial cells are crushed using the crushing beads contained in the Bead tubes and DNA is extracted
  • It contains a unique additive that inhibits the adsorption of DNA to soil particles, enabling the recovery of DNA at a high yield.
  • Since the resulting DNA solution contains few inhibitory substances, the undiluted solution can be used directly for PCR.
  • The use of magnetic beads for DNA purification makes the operation simple and automated.
  • Even when microbial cells are disrupted with a vortex mixer, almost the same results are obtained as when using a bead beader.
  • Since organic solvents such as phenol and chloroform are not used, it is safe to use.

How to Use


Cell Disruption (Bead Beating)
(1) Add 0.5 g of soil sample, Extraction buffer, and Lysis solution to a Bead tube, and perform bead beating*3 (30-45 sec).
(2) Centrifuge (5 min) and collect supernatant.
Deproteinization
(3) Add PP solution, centrifuge (5 min), and collect supernatant.
Magnetic Bead Purification
(4) Add MBs solution and Binding solution, stir (2 min), collect magnetization, and discard supernatant.
(5) Add washing solution, stir, collect magnetization, and discard supernatant.
(6) Add 70% ethanol, stir, collect, and discard supernatant.
DNA Elution
(7) After air-drying, add elution buffer, and warm at 65°C for 5-10 min.
(8) Magnetization, supernatant collection → total DNA.

*3 Please use a bead type crusher (sample/cell crusher) for bead beating. You can also use a vortex mixer with a vortex adapter (Vortex Adapter for Vortex-Genie 2 (Qiagen)).

Comparison of DNA Extraction Efficiency

Extrap example of use

Comparison tests with kits from other companies were conducted for soils that are generally difficult to extract. DNA was extracted from seven soils using the Extrap Soil DNA Kit Plus Ver2 and three kits from other companies. As a result, DNA extraction values equivalent to or higher than those of other companies were obtained for all soils. The Extrap Soil DNA Kit Plus Ver2 can recover DNA from not only soil but also a wide variety of environmental samples with high yields.

Assay value of undiluted DNA sample ÷ Assay value of 100 fold diluted DNA sample
If there is no PCR inhibition, it is close to 1, and the smaller the value, the greater the PCR inhibition. 0.5 or less is shown in red letters. From the experimental results, it was confirmed that the removal effect of PCR inhibitors by Extrap was equal to or greater than that of kits from other companies.
