What is S1P

Sphingosine-1-phosphate (S1P) is a lipid signaling molecule that regulates various physiological functions via its receptors, including the trafficking of cancer-related immune cells, vascular permeability, and angiogenesis. In research targeting the S1P receptor S1PR3, X-ray crystallography has elucidated the three-dimensional structure recognized by S1P by focusing on its localization in the cell membrane and binding to the extracellular domain.

What is GPR87

In recent years, GPR87 has emerged as a promising therapeutic target and cancer marker, as it is expressed in various types of cancer while showing minimal expression in normal tissues 1. Research using humanized antibodies, derived from anti-GPR87 mouse monoclonal antibodies, has demonstrated the efficacy of GPR87-targeted near-infrared photoimmunotherapy in models of lung cancer and malignant pleural mesothelioma 2.

Example of Use

EDG3/S1PR3 antibody (PP-AS55-0C)

Flow Cytometry ; Anti human EDG3/S1PR3 mouse monoclonal antibody/ AS55

Two experimental groups were prepared using the human breast cancer cell line Cama-1: an S1PR3-targeted siRNA group (siRNA-S1PR3 group) and a negative control siRNA group. Knockdown efficiency was evaluated by qPCR, with relative expression levels calculated using the 2-ΔΔCt method. The results showed that S1PR3 expression was significantly reduced in the siRNA-S1PR3 group compared to the negative control group. Additionally, the specificity of antibody staining was assessed via flow cytometry.

GPR87 antibody (PP-C0804-0C, PP-C0812-0C)

Flow Cytometry GPR87 antibody

GPR87-overexpressing cells (2×105 cells) were incubated with the primary antibody at a final concentration of 1 μg/mL for 1 hour. After washing, PE-labeled anti-mouse IgG (secondary antibody: Jackson ImmunoResearch Laboratories, 1:200) was added, and the cells were incubated for 1 hour at 4°C in the dark. After a final wash, the cells were analyzed using a flow cytometer. A peak shift was confirmed at the final concentration of 1 μg/mL.
