This kit is designed for constructing protein measurement systems. It operates on the principle of the OpenGUS Immunoassay using a mutant variant of β-glucuronidase (GUS). Compared to traditional ELISA, this method offers simpler operations and requires a smaller amount of antibodies. It also provides higher quantitativeness in less time than Western blotting.

Features

  • No Immobilization or Washing Required: Eliminates the need to immobilize antibodies onto plates or wash the wells.

Principle

ONEPot Kit Principle

E. coli β-glucuronidase (GUS) exhibits enzymatic activity by forming a tetramer. The mutant GUS used in this product is fused with an antibody-binding domain; it forms dimers naturally but rarely forms tetramers on its own.
When the target antigen is present, the antigen-antibody reactions link the mutant GUS dimers together, forcing the formation of an active tetramer. The resulting enzymatic activity produces a signal dependent on the antigen concentration.

Protocol Overview

  1. Mix the measurement sample (or purified antigen for the standard curve), the two types of antibodies, and the enzyme included in the kit.
  2. Incubate at room temperature for 60 minutes.
  3. Add the substrate included in the kit.
  4. Incubate at room temperature for 60 minutes.
  5. Measure the fluorescence or absorbance using a plate reader.

Application Example

Measurement of Lactoferrin Concentration in Saliva and Tears

Lactoferrin concentrations in saliva and tears were measured using this product and compared with the results obtained from a commercially available Lactoferrin ELISA kit.

Precautions

  • Antigen Concentration: If the target antigen is a protein, it is recommended to use samples containing (3 μg/ml) or more.
  • Antibody Compatibility: Even if an antibody pair works successfully in a standard sandwich ELISA, it may not always be compatible with this kit.
  • Sample Composition: Components within the sample solution can significantly influence signal intensity.
  • Incompatible Samples: Samples containing high amounts of immunoglobulins, such as human serum, plasma, or whole blood, cannot be used due to high background interference.
