These antibodies specifically recognize the drug payload of an Antibody-Drug Conjugate (ADC), enabling detection and quantification via ELISA and immunohistochemistry (IHC) — methods that previously required expensive equipment like HPLC-MS/MS. They are widely applicable, ranging from early-stage development screening to preclinical pharmacokinetics (PK) analysis and tissue distribution studies.
What is ADC Payloads
DM1
A cytotoxic payload utilized in antibody-drug conjugates (ADCs) that acts by inhibiting microtubule polymerization. A prominent example is trastuzumab emtansine (T-DM1, trade name: Kadcyla). T-DM1 is composed of the anti-HER2 antibody trastuzumab, a linker, and DM1, specifically targeting HER2-positive breast cancer.
MMAE
A representative cytotoxic payload utilized in ADCs that induces mitotic arrest and cancer cell apoptosis by inhibiting microtubule formation. Additionally, the liberated MMAE can exert a bystander killing effect on adjacent cell populations.
DXd/Exatecan
A cytotoxic payload utilized in ADCs, functioning as an exatecan-derived topoisomerase I inhibitor. It mediates intracellular cytotoxicity and demonstrates a bystander effect by diffusing into and eliminating surrounding cells.
