Nipp also offers Brightase-TH, a recombinant thermolysin expressed in Brevibacillus choshinensis. For optimal results, a set product featuring both Brightase-C and Brightase-TH is available. This allows you to adjust the ratio of the two enzymes to achieve the most effective cell isolation according to the specific tissue type being processed.
Features

*Since Brightase-C does not contain neutral protease, we highly recommend using it in combination with Brightase-TH when isolating various cells from biological tissues, such as in islet isolation or adipose-derived stem cell extraction.
Technical Data
Expression System
The Brevibacillus expression system used to produce Brightase-C (this product) offers key advantages: it is well-suited for extracellular protein expression, enables target protein purification directly from the culture supernatant, and is low in endotoxins as well as free of animal-derived components and antibiotics (neomycin).
Enzyme Stability
The 74 kDa full-length enzyme containing the PPC (pre-peptidase C-terminal) domain*1 undergoes autolysis, which cleaves it into a 62 kDa form and reduces its activity. In contrast, the 62 kDa enzyme possessing only the catalytic domain remains stable without undergoing autolysis, even after being incubated at 37°C for one day. Consequently, the 62 kDa enzyme (Brightase-C, this product) is the ideal choice for a collagenase product due to its superior enzymatic activity stability.
*1 PPC domain: Confirmed to function as a CBD (collagen-binding domain).
