These are pET expression vectors that uses the highly efficient T7 promoter to express the desired protein in E. coli. There are high copy number vectors, medium copy number vectors, and vectors whose expression can be controlled by LacI, each containing a drug resistance gene for ampicillin or kanamycin.
pET System
The T7 expression system is a powerful expression system using a pET expression vector containing the T7 promoter and a lysogen of bacteriophage λDE3, such as the BL21 (DE3) E. coli strain, into which the T7 RNA polymerase gene is integrated.
The expression of the T7 RNA polymerase gene under the control of the lac UV5 promoter in the BL21 (DE3) E. coli genome is induced by the addition of isopropyl-1 thio-β-D-galactopyranoside (IPTG). The expressed T7 RNA polymerase recognizes the T7 promoter of the pET vector and strongly expresses the target protein sequence introduced downstream of the T7 promoter.
Feartures
- All six vectors share a common multi-cloning site. This makes it easy to subclone from one series to another.
- Because of the small size of the vectors, high-efficiency transformation can be expected even with large inserts.
- Proteins fused with an N-terminal His tag can be expressed. Untagged proteins can also be expressed by inserting the target gene at the Nde I site in the cloning site.

* When the effect of the protein to be expressed on the host E. coli is not clear, or when the choice of vector is difficult, try pETBA. If you want to suppress the expression in the uninduced state, use pETIA. When the protein to be expressed has little effect on the host E. coli, use pETUA, which has a large plasmid copy number, has the advantage of making miniprep and sequencing easier.
* pETUK, pETBK, and pETIK, which contain the kanamycin resistance gene, have the same characteristics as pETUA, pETBA, and pETIA, respectively. These vectors are useful for subcloning from ampicillin-resistant cloning vectors.
