What is HAMA?
HAMA is said to be present in a few percent of healthy individuals and is one of the causes of false positives in immunoassays. Therefore, HAMA inhibitors are generally added to immunoassays in advance to avoid interference by HAMA. In clinical practice, when false positives due to HAMA are suspected, the addition of HAMA inhibitors may enable differentiation.
Principle
In sandwich immunoassays using mice (①), the presence of HAMA in samples may cause a cross-linking reaction (non-specific reaction) between the solid phase antibody and the labeled antibody, resulting in a false-positive result (②). In this case, adding an HAMA inhibitor can block HAMA and suppress the non-specific reaction (③).

* The effect of THBR1-AS/THBR2 varies depending on the measurement system and non-specific sample. When using the product, it is recommended to examine the concentration of THBR1-AS/THBR2 in each measurement system.
* THBR1-AS (#2ATHBR1) can also be applied to coagulation methods.
Example of Use
In a 2-step sandwich ELISA using mouse IgG1 antibody as a solid phase antibody and labeled antibody, THBR1-AS/THBR2 was added to the sample diluent and labeled antibody diluent. In this case, the addition of THBR1-AS: 0.1-1% and THBR2: 10-100 μg/ml almost completely inhibited the non-specific reaction induced by HAMA.
