HB S Antigen Quantitative ELISA (#BCL-SHP-21)

These kits are designed to quantify the S, Pre-S1, and Pre-S2 regions of the Hepatitis B Virus surface antigen (HBsAg). Each kit specifically detects its respective region. Highly reproducible quantification is achieved using the included recombinant standard antigens. For the Pre-S1 region, a dedicated kit is available to detect Genotype D, which was difficult to measure with conventional products. These kits are highly useful for studying HBV infection mechanisms and developing therapeutic drugs.

What is HBsAg

HBsAg consists of three forms: S-antigen, M-antigen, and L-antigen.

  • L-antigen (Full-length): Composed of Pre-S1, Pre-S2, and S domains.
  • M-antigen: Composed of Pre-S2 and S domains.
  • S-antigen: Composed of the S domain only.
Each region plays a critical role in HBV infection. The Pre-S1 region is thought to be important for recognition of hepatocytes, which are host cells, and the Pre-S2 and S regions are thought to be important for viral entry into hepatocytes.

  • Two kit types are available: a rapid assay kit with relatively lower sensitivity for genotype D (#BCL-S1HP-01), and a specialized kit capable of detecting genotype D (#BCL-S1D-01).

Q1: What are the storage conditions and shelf life for this kit?

A1: All reagents included in this kit should be stored at 4°C. The antigen quantitative ELISA kits remain stable for up to 6 months.

Q2: How long does the assay take?

A2: The approximate incubation times required for the assay (excluding the time needed for reagent preparation, sampling, washing steps, and absorbance measurement) are as follows. For further details, please refer to the product manual.

· For #BCL-SHP-21, #BCL-S1HP-01, and #BCL-S2HP-01:
① Capture antibody and detection antibody reaction: 1 hour → ② Color development: 30 minutes

· For #BCL-S1D-01 (Genotype D):
① Capture antibody reaction: 3 hours → ② Detection antibody reaction: 3 hours → ③ Color development: 30 minutes

Q3: What do the measurement units mean?

A3: There is no international standard unit for Pre-S1 and Pre-S2 antigens. In these kits, 1 nUnit is defined based on Beacle, Inc.’s proprietary standard, where 1 nUnit corresponds to the Pre-S1 or Pre-S2 antigen titer contained in 1 ng of Beacle's standard antigen.
For the HB S Antigen Quantitative Kit, 1 nUnit is approximately equivalent to 0.5 to 1.0 IU according to the WHO international standard.

Q4: What is used as the standard antigen?

A4: The standard antigens provided are recombinant proteins produced in yeast or E. coli. There is no risk of biohazardous viral infection.

Q5: I do not have data analysis software. How should I calculate the measurement values?

A5: As described in each kit's manual, the concentration should be calculated by fitting the absorbance data to a 4-parameter logistic (4PL) regression model. Although most microplate readers come with dedicated software, if you do not have access to it, you can download our complimentary Excel-based calculation program below.
