複数のサイトカインを同時検出 Proteome Profiler Cytokine Antibody Array Kit

単一試料中に存在する多数のサイトカインを、同時に検出できる抗体アレイキットです。従来の免疫沈降／ウエスタンブロッティングによる方法よりも簡便で、短時間により多くのサイトカインの解析を行えます。各キットには特異性の高い抗体を 2 か所ずつスポットしたニトロセルロースメンブレンアレイが 4 枚、各種バッファーおよびビオチン標識した検出抗体が含まれています。

36 種類の抗サイトカイン抗体をスポットしたアレイが含まれています。細胞培養上清、血清、血漿（ EDTA, ヘパリン処理）、尿、唾液、母乳中の各種ヒトサイトカインを同時に検出できます。

Figure 1A: THP-1 human acute monocytic leukemia cells were untreated or treated with 1 μg/mL LPS for 4 hours, then 5mM Nigericin (Tocris, Catalog # 4312) for 1 hour (500 μL of cell culture supernate, 10 minute exposure).

Figure 1B: SW480 human colorectal adenocarcinoma cells were untreated or treated with 100 ng/mL LPS for 24 hours (500 μL of cell culture supernate, 10 minute exposure).

40 種類の抗サイトカイン抗体をスポットしたアレイが含まれています。細胞培養上清、細胞ライセート、血清、血漿（ EDTA, ヘパリン処理）中の各種マウスサイトカインを同時に検出できます。

■使用例

Mouse splenocytes treated with 100 ng/mL LPS for 20 hours were mixed with a cocktail of biotinylated detection antibodies, and then incubated with the Mouse Cytokine Array. The array was then incubated with streptavidin-horseradish peroxidase followed by chemiluminescent detection. After detection, the array data were quantitated to generate a protein profile (histogram). The table shows the analytes detected and their location on the membrane.
