Q-1. 公称粒子径と測定された平均粒子径の関係は?

A-1. Micromod社の製品説明や技術データシートに記載されている粒子径はすべて公称値であり、測定された平均粒子径とは異なる場合があります。直径<2μmの粒子径は、Zetasizer Nano ZS 90(Malvern Instr社)を用いた動的光散乱(DLS)によって測定されます。DLSは、光子相関分光法(PCS)としても知られています。≧2μmの粒子径は、コールター®マルチサイザーⅢで測定します。粒子径と各粒子タイプの粒度分布の仕様は、ご要望に応じて提供しています。ご希望の場合は、テクニカルサポート部 試薬担当へお問い合わせ下さい。


Q-2. ミクロスフェア粒子のサイズはどのように測定しますか?

A-2. インピーダンス法を用いたマルチチャンネル粒度分布測定装置で、直径≧2μmの粒子径を測定します。この方法は、粒子が測定ゾーンを通過するときのインピーダンス*の変化に基づいています。この変化は、粒子の体積に比例します。

Q-3. 変動係数(Coefficient of Variation、%CV)とは何ですか?

A-3. 変動係数(CV)とは、一般的にビーズ集団のサイズのばらつきを意味します。CVは、平均値に対するばらつきの度合いをパーセントで表したものです。CVは、標準偏差を平均粒子径で割ることで得られます(例:%CV = 100% × 標準偏差/平均粒径)。Micromod社の直径2~10μmのmicromer粒子のCV値は≦5%です。


Q-4. ナノ粒子の大きさはどのように測定しますか?

A-4. 一般的に、直径が<2μmの粒径は、Zetasizer Nano ZS 90(Malvern Instr.社)を用いた動的光散乱(DLS)によって測定されます。DLSは光子相関分光法(PCS)としても知られています。
Zetasizerは、まず動的光散乱(DLS)を用いて試料中の粒子のブラウン運動を測定し、次に確立された理論を用いて粒径を決定します。PCS測定のデータ分析には、次の4つの異なるタイプがあります。

  • キュムラント解析
  • 多重指数解析
  • CONTIN分析
  • 非負の最小二乗法
Z平均サイズは、この技術によって生成される最も重要で安定した数値です。これは、品質管理のために数値が必要な場合に使用するサイズで、キュムラント分析によって得られます。この分析では、サイズの平均値と多分散性指数(PDI)と呼ばれる幅のパラメータの2つの値しか得られません。この平均サイズは強度平均であり、信号強度から直接計算されます。Micromod社では蓄積した経験を基にナノ粒子の評価には様々なタイプの分析法を使用しています。特に、蛍光ナノ粒子や磁性ナノ粒子の異なる散乱特性は、分析方法の最適な調整が必要です。


Q-5. 多分散指数(PDI、Polydispersity Index)とは何ですか?

A-5. ナノ粒子の粒度分布は、光子相関分光法(PCS)で測定されます。粒度分布の質は、多分散指数(PDI)で示されます。この値は、測定された自己相関関数と、数学的に適合された相関関数との偏差として計算されます。粒度分布が小さいナノ粒子では<0.2、粒度分布が広いナノ粒子では、0.2~0.5の間です。PDIの値が>0.5の場合は、測定値を分析できないことを示しています。


Q-6. 粒子の表面電荷はどのようにして測定していますか?

A-6. Zetasizer Nano ZS 90(Malvern Instr.社)を使用して、粒子のゼータ電位を測定します。ゼータ電位は、電気泳動とレーザードップラー流速計(レーザードップラー電気泳動法と呼ばれることもあります)の測定手法を組み合わせて測定されます。この方法では、電界が印加されたときに液体中で粒子がどれだけ速く移動するか、つまり速度を測定します。


Q-7. 磁性粒子の懸濁液を大量に使用する場合、どの磁気分離装置がお勧めですか?

A-7. 1Lまでの容量の場合は、SEPMAG Q1L(www.sepmag.eu)をお勧めします(当社非取扱品です)。例えば、nanomag-D、500 nmの粒子(V=600 ml、c=10 mg/ml, 製品コード:09-00-502)は、SEPMAG Q1Lを用いて約30分で分離しています。


Q-8. 表面にアミノ基を持つシリカ粒子のゼータ電位が負になるのはなぜですか?

A-8. シリカ粒子のアミノ官能化は、シリカ粒子表面のSi-OH基を3-(アミノプロピル)トリエトキシシランで重縮合することで行われます。NH2基の他にも、粒子表面には酸性のSi-OH基が多数存在しており、これがアミノ基標識シリカ粒子の全体的な負のゼータ電位の原因となります。直径1μmの普通のシリカ粒子とアミノ基標識シリカ粒子のゼータ電位-pH関数を比較すると、普通のシリカ粒子のゼータ電位よりアミノ基標識シリカ粒子では正の方向にシフトしています。アミノ基標識シリカ粒子は、pH<4で正のゼータ電位を有し、pH>4で負のゼータ電位を有します。
