 | Detection of Phospho-IFN-alpha / beta R1 (S535/S539) by Western Blot Western blot of immunoprecipitates from HEK293 cells transfected with 1) Mock, 2) IFN-alpha R1 wild type, and 3) IFN-alpha R1 S535A and S539A mutants showing specific immunolabeling of the ~110 kDa to ~130 kDa IFN-alpha R1 wild type. The immunolabeling is absent in the IFN-alpha R1 S535 and S539 mutants (Control). The immunolableing is blocked by the phosphopeptide (Phos) used as the antigen but not by the corresponding dephosphopeptide (Dephos). |

These cytokines produce their effects by binding to the Type I IFN Receptor. This receptor contains two subunits; IFNAR1/ alpha 1 and IFNAR2/ alpha 2 Down regulation of this receptor plays a key role in determining the magnitude and duration of cytokine signaling. This downregulation is thought to be influenced by phosphorylation of S535 and S539 in IFN-alpha R1 (3). |
