Figure 1. Western blot of GFP recombinant protein detected with GeneTex polyclonal anti-GFP antibody. Lane 1 shows detection of a 33 kDa band corresponding to a GFP containing recombinant protein (arrowhead) expressed in HeLa cells. Lane 2 shows no staining of a mock transfected HeLa cell lysate. After blocking the membrane was probed with the primary antibody diluted to 1 μg/ml for 1 h at room temperature followed by washes and reaction with a 1:2,500 dilution of donkey anti-Goat IgG [H&L].
Sf-1+ neurons and their processes of the ventromedial nucleus of the hypothalamus in Mus musculus (coronal view, 20X magnification). GeneTex polyclonal goat anti-GFP was used at 1:500 dilution in free floating imunnohistochemistry to detect eGFP. Fluorchrome conjugated anti-goat IgG secondary antibody was used for detection at 1:500. Sections were counterstained with DAPI. Personal Communication, Daniel A. Tissue: E5.5 Hex-GFP transgenic mouse embryo. Primary antibody: Goat anti-GFP was used at 1:500 dilution. Secondary antibody: Fluorchrome conjugated Anti-goat IgG secondary antibody at 1:10,000 for 45 min at RT. Staining: GFP as green fluorescent signal with DAPI blue counterstain.

TARGET

Background The jellyfish Aequorea victoria contains green fluorescent protein (GFP) that emits light in the bioluminescence reaction of the animal. GFP has been used widely as a reporter protein for gene expression in eukaryotic and prokaryotic organisms, and as a protein tag in cell culture and in multicellular organisms. As a fusion tag, GFP can be used to localize proteins, to study their movement or to research the dynamics of the subcellular compartments where these proteins are targeted. GFP technology has revealed considerable new insights into the physiological activities of living cells. GFP is a 27 kDa monomeric protein, which autocatalytically forms a fluorescent pigment. The wild type protein absorbs blue light (maximally at 395nm) and emits green light (peak emission 508nm) in the absence of additional proteins, substrates, or co-factors. GFP fluorescence is stable, species independent and is suitable for a variety of applications. GFP has been used extensively as a fluorescent tag to monitor gene expression and protein localization. Moreover, other applications for GFP include its use in assessing protein protein interactions in the yeast two hybrid system, and in measuring distances between proteins in fluorescence energy transfer (FRET) experiments.
