Background: SPRY1

SPRY1 (sprouty homolog 1) is a 34-38 kDa member of the sprouty family of proteins. It is widely expressed, being found in multiple embryonic and adult tissues. SPRY1 is considered a negative regulator of cellular signaling. In particular, it appears to both inhibit MAP kinase signaling following RTK activation, and block TCR signaling following antigen activation. It interacts with a number of molecules, including PLC-γ1, LAT, CBL, caveolin-1 and SPRY2. Human SPRY1 is 319 amino acids (aa) in length. It contains one CBL-TKB binding site (aa 51-57) that is phosphorylated at Tyr-53, a Ser-rich region (aa 112-131), and a Cys-rich domain (aa 181-306) that mediates intracellular translocation. SPRY1 undergoes serine phosphorylation, ubiquitination and palmitoylation, the latter which induces SPRY1 to associate with cell membranes. Over aa 1-178, human SPRY1 shares 76% aa identity with mouse SPRY1.

■Western Blot

Detection of Human SPRY1 by Western Blot.

Western blot shows lysates of 293T human embryonic kidney cell line and LNCaP human prostate cancer cell line. PVDF Membrane was probed with 0.5 µg/mL of Human SPRY1 Monoclonal Antibody (# MAB6097) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (# HAF007). A specific band was detected for SPRY1 at approximately 37 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Background: SPRY2

SPRY2 (sprouty homolog 2) is a 35 kDa member of the sprouty family of proteins. It is widely expressed in embryonic and adult tissues, including endothelium. SPRY2 is considered a negative regulator of Ras/ERK signaling. For example, it appears to form a trimeric complex with PKCδ and PKD1, blocking ERK phosphorylation. It also interacts with a number of other molecules, including CBL and SIAH2, which likely limit its availability. Human SPRY2 is 315 amino acids (aa) in length and contains one CBL-TKB binding site (aa 53 - 59) that is phosphorylated at Tyr55, a Ser-rich region (aa 125 - 131), a Cys-rich domain (aa 178 - 301) and a PxxPxR motif that mediates ERK inhibitory activity. Over aa 1 - 175, human SPRY2 shares 94% aa identity with mouse SPRY2.

Western blot shows lysates of SH‑SY5Y human neuroblastoma cell line, PC‑3 human prostate cancer cell line, and C2C12 mouse myoblast cell line. PVDF Membrane was probed with 1 µg/mL of Goat Anti-Human/Mouse SPRY2 Antigen Affinity-purified Polyclonal Antibody (# AF6157) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (# HAF109). A specific band was detected for SPRY2 at approximately 35 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Background: SPRY3

SPRY3 (sprouty homolog 3) is a member of the sprouty family of proteins. SPRY3 appears to modulate Ras/MAPK pathway signaling following RTK activation. Human SPRY3 is 288 amino acids (aa) in length and contains one SH2binding domain (aa 25-30), a PEST sequence (aa 82-93), and a Cysrich domain (aa 147-270). SPRY3 forms homoand hetero-oligomers with other SPRY family molecules. It also undergoes phosphorylation, ubiquitination and almitoylation, the latter of which induces SPRY3 to associate with cell membranes.

Detection of Human SPRY3 by Western Blot.

Western blot shows lysates of HepG2 human hepatocellular carcinoma cell line, MCF‑7 human breast cancer cell line, and Caco‑2 human colorectal adenocarcinoma cell line. PVDF membrane was probed with 1 µg/mL of Human SPRY3 Antigen Affinity-purified Polyclonal Antibody (# AF5735) followed by HRP-conjugated Anti‑Sheep IgG Secondary Antibody (# HAF016). A specific band was detected for SPRY3 at approximately 42 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Background: SPRY4

SPRY4 (sprouty 4) is a member of the sprouty family of proteins. It blocks tyrosine kinase growth factor activation of the MAPK signaling pathway, possibly through an interaction with either Raf1 or TESK1. Human SPRY4 is 322 amino acids (aa) in length. It contains three SH3-binding domains (aa 3-9; 25-31; 168-174), an NLS (aa 87-93), a PEST sequence (aa 118-133), and a Cys-rich/Zn++-finger domain that binds both TESK1 and Raf1 (aa 182-298). There are two variant isoforms. One is a short form that shows an alternate start site at Met24, while a second shows a 10 aa substitution for aa 120-129, followed by a premature truncation. Over aa 1-177, human SPRY4 is 89% aa identical to canine SPRY4. Mouse SPRY4 is orthologous to the human short form, and shares 90% aa identity with human SPRY4.
