R&D Systems now offers a high-purity form of recombinant low-density lipoprotein receptor (LDL R). Recombinant Human LDL R (High Purity) (Catalog # 2148-HP) contains 90% or greater full-length LDL R extracellular domain, as compared to the up to 50% truncated forms in R&D Systems standard Recombinant Human LDL R (Catalog # 2148-LD ) or up to 100% truncation observed in competitors' products. The truncated recombinant forms of the LDL R extracellular domain lack several repeats of Class A LDL domains near the N-terminus. These domains are responsible for binding to ApoB-100 and ApoE, the apolipoproteins of low- and very low-density lipoproteins (LDL and VLDL), respectively. Endogenous active LDL R extracellular domains of similar molecular weight to Recombinant Human LDL R (High Purity), can be released from cells by treatment with phorbol esters.

Purity of Recombinant Human LDL R and its Competitive Binding to Immobilized Human LDL. A. Purity is highlighted by silver staining of proteins resolved by SDS-PAGE under reducing conditions, showing Recombinant Human LDL R (Catalog # 2148-LD, center lane) and Recombinant Human LDL R (High Purity) (Catalog # 2148-HP, right lane) as compared to molecular weight markers (left lane). B. Unlabeled Recombinant Human LDL R (High Purity) (Catalog # 2148-HP, red) or Recombinant Human LDL R (Catalog # 2148-LD , green) was mixed with 0.5 µg/mL of biotinylated Recombinant Human LDL R. This mixture was then incubated for 2-3 hours in a 96-well plate immobilized with human LDL at 1 µg/mL (100 µL/well). Wells were washed and biotinylated LDL R bound to the plate was visualized with Streptavidin-HRP.
