Modelling disease in 2D progenitor cell cultures: From bench to bedside of Pemphigus vulgaris patients Key Topics: Pemphigus vulgaris is a life-threatening autoimmune disease targeting desmosomal cadherins at the adhesive interface between stem and progenitor cells of stratified epithelia and the hair follicle bulge, one of the best-studied stem cell niches in the mammalian body. This presentation highlights how accurate disease modelling in 2D cultures led to an entirely novel, consensus-backed concept of Pemphigus vulgaris pathophysiology. Two major benefits concerning clinical applications and regenerative medicine emanate from these advances; i) novel therapies for pemphigus vulgaris patients and ii) the understanding of an unfathomed role of a desmosomal cadherins in niche homeostasis and a self-organizing repair program in the injured stem cell niche. |
