Netrin-1はヒトおよびマウス体細胞のアポトーシスを阻害することにより、iPS細胞の形成、およびリプログラミング効率を改善します。またNetrin-1は中枢神経系の交連軸索と末梢運動軸索の伸長を制御します。

Netrin-1(ネトリン-1)とは
Netrin-1 seems to be the 5th Element of classical iPS cell factors. Netrin-1 induces pluripotent stem cell (iPS) formation and improves reprogramming efficiency of human and mouse somatic cells by inhibiting apoptosis mediated by the receptors DCC or UNC5B (at 150ng /ml). Together with Netrin-1 the original factors, Oct4, Sox2, Klf4 and c-Myc can produce 15 times more induced pluripotent stems cells (iPSCs) under specific culture conditions. This treatment did not affect the quality of cell reprogramming. Further research is ongoing for a better understanding of the mode of action of this protein in stem cell physiology to accelerate the development of clinically useful iPS cells.

使用例
Netrin-1 (human):Fc (human) (rec.) (#AG-40B-0075) triggers a DCC-dependent phosphorylation of ERK1/2.Method: HEK 293 cells (Control) or HEK 293 expressing the netrin-1 receptor DCC were incubated with netrin-1 (human):Fc (human) (rec.) (5 nM) for the indicated time points (shown in minutes). Antibodies against DCC, total ERK1/2 or phospho-ERK1/2 were used and visualized with a chemiluminescence detection system.
Netrin-1 (human):Fc (human) (rec.) (#AG-40B-0075) induces outgrowth of the commisural axon.Method: Dorsal spinal cords were dissected out from E13 rat embryos and cultured in collagen matrix in the presence or absence of netrin-1 (250ng/ml). Axons were then stained with an anti-β-tubulin antibody.
