唾液中ゲノムDNAの保存・ハイスループット解析に最適なキット DNAgard Saliva HT

唾液中のゲノムDNAを室温条件下で長期間安定に保存できるキットです。ゲノムDNA抽出用の唾液試料の輸送や長期保存、自動化システムによるハイスループット解析に最適です。

Saliva samples from 5 donors collected individually into conical tubes were aliquoted into tubes containing DNAgard Saliva HT solution (DGS HT), Competitor’s solution (CO), or in a conical tube with non–protective control solution (NP). All samples were stored at 45℃ for 5 weeks and moved to 60℃ for an additional 9 days, equivalent to at least 10 months at 22℃ calculated using the Arrhenius equation.

Preserved DNA integrity after10 months at room temperature

Agarose gel of DNA purified from saliva of five donors stored using Competitor’s solution, DNAgard Saliva HT, or non-protected saliva samples. DNA was visualized on a 1% agarose gel stained with ethidium bromide.

High DNA yield after 10 months at room temperature

dsDNA yield of DNA isolated from saliva stored using Competitor’s solution, DNAgard Saliva HT, or non-protected saliva samples. DNA concentration was measured using Quant-iT PicoGreen dsDNA Assay.

Detection of targets in PCR assays

qPCR of human DNA stored using Competitor’s solution, DNAgard Saliva HT, or nonprotected saliva samples. Samples were amplified in duplicate using iQ SYBR Green Supermix (Bio-Rad), on a CFX96 Real-Time PCR Instrument (Bio-Rad) using primers specific to the human RNase P gene.
