Flow Cytometry: HLA-DR Antibody (L243) [PerCP/Cy5.5] [NBP2-27183] - Cell surface analysis of HLA-DR expression by 5x10^5 human PBMC using 10 ul (0.25 ug) of this antibody. Cell surface flow kit was used for this test. Left panel shows the monocyte gate, right panel shows the lymphocyte gate. In general, monocyte population gives background with mouse IgG2a antibody.

Background

L243 antibody recognizes MHC class II molecules expressed on antigen presenting cells, such as dendritic cells, macrophages, B cells, monocytes and activated T cells. MHC class II molecules are hetero-dimeric ( (36kD) and (27kD) subunits) proteins, which bind peptides derived from pathogens and present to CD4 T cells, to initiate and activate adaptive immune responsesThis antibody reacts with both peptide free (empty) and bound conformations of class II molecules, with multiple epitopes on dimeric complex. Antibody can be used in monitoring dendritic cell differentiation and maturation and also in blocking experiments to monitor the MHC II mediated antigen presentationThis antibody is a nice tool to study both structural and functional studies of MHC class II molecules, besides flow (cytometry) based studies on cells expressing the molecules.
