R&D Systems offers a Mouse Anti-Human IDO Monoclonal Antibody (Catalog # MAB6030 ). Indoleamine 2,3-dioxygenase (IDO) is a heme-containing intracellular dioxygenase that catalyzes the degradation of L-tryptophan to N-formylkynurenine, the first and rate-limiting step of the L-kynurenine pathway. IDO is expressed in dendritic cells, macrophages, microglia, eosinophils, fibroblasts, endothelial cells, and most tumor cells. In immune cells, its expression is mainly induced by cytokines such as IFN-alpha, IFN-beta, IFN-gamma and IL-10. The L-kynurenine pathway has both antimicrobial and immunosuppressive effects since depletion of the essential amino acid tryptophan can arrest the growth of microbes and T cells. As a result, IDO inhibits microbial activity in inflammatory environments, and has been shown to induce immunosuppression during infection, pregnancy, transplantation, autoimmunity, and neoplasia.

IDO in A431 Cells. IDO was detected in immersion-fixed A431 human epithelial carcinoma cells, stimulated with Recombinant Human IFN-gamma (Catalog # 285-IF), using a Mouse Anti-Human Indoleamine 2,3-dioxygenase/IDO Monoclonal Antibody (Catalog # MAB6030). Cells were stained using the NorthernLights™ 557-conjugated Donkey Anti-Mouse IgG Secondary Antibody (Catalog # NL007 ; red) and counterstained with DAPI (blue).
