R&D Systems offers Human StemXVivo Serum-Free MSC Expansion Media (#CCM014). This media is specially formulated and optimized for the maintenance and expansion of purified human mesenchymal stem cells (MSCs). MSCs are phenotypically characterized as Endoglin/CD105+, CD90/Thy1+, CD73/5' Nucleotidase+, CD44+, CD45-, CD34 -, and Integrin alpha M/CD11b- cells. These cells have both the capacity to self renew and to differentiate into multiple cell types, including adipocytes, chondrocytes, and osteocytes. The Serum-free MSC Expansion Media is a ready-to-use formulation for culturing human MSCs on plates coated with extracellular matrix proteins. Alternatively, the media can be used with cytokine/growth factor supplements for additional cell culture applications.

A. Morphology of human MSCs cultured with Human StemXVivo Serum-Free Mesenchymal Stem Cell Expansion Media (#CCM014). B. Expanded MSCs analyzed by flow cytometry using PE-conjugated Anti-Endoglin/CD105 (#FAB10971P), Anti-CD90/Thy1 (#FAB2067P), Anti-CD45 (#FAB1430P), or Anti-Integrin alpha M/CD11b (#FAB16991P; open histograms) or isotype controls (filled histograms). C. Human MSCs were grown in Human StemXVivo Serum-Free MSC Expansion Media and differentiated using the Human Mesenchymal Stem Cell Functional Identification Kit (#SC006). Adipocytes were stained with Anti- FABP4 (#AF1443; left panel), osteocytes with Anti-Osteocalcin (#MAB1419; middle panel), and chondrocytes with Anti-Human Aggrecan (#AF1220; right panel). The nuclei were counterstained with DAPI (blue).
