R&D Systems offers a Chicken Anti-Human/Mouse/Rat Neurofascin Antigen Affinity-purified Polyclonal Antibody (AF3235). Neurofascin is a type I transmembrane glycoprotein that belongs to the L1CAM family of cell adhesion proteins. The antibody recognizes two isoforms that result from alternative splicing. Neurofascin 155 (NF155) is transiently expressed by oligodendrocytes at the onset of axon myelination and is stabilized in paranodal oligodendroglia lipid raft domains by dimerization of its cytoplasmic domains and association with ankyrin. NF155 is involved in establishing the paranodal axoglial junctions that flank nodes of Ranvier. Neurofascin 186 (NF186) is neuronally expressed in nodes of Ranvier and is essential for clustering of sodium channels at nascent nodes.

Detection of Neurofascin by Immunohistochemistry and Western Blot.
A. Neurofascin was detected in perfusion-fixed, frozen sections of rat dorsal root ganglion using a Chicken Anti-Human/Mouse/Rat Neurofascin Antigen Affinity-purified Polyclonal Antibody (#AF3235). The tissue was stained using the NorthernLights™ 557-conjugated Anti-Chicken IgY Secondary Antibody (#NL016; yellow) and counterstained with DAPI. Specific staining was localized to neuronal cell bodies and Schwann cells (perinodal regions).
B. Neurofascin was also detected by Western blot in samples from rat, mouse, and human brain.
