R&D Systems offers a Glycosyltransferase Activity Kit (#EA001). This kit provides a universal, non-radioactive assay for determining the enzymatic activity of glycosyltransferases that use diphosphonucleotide sugars as donor substrates. The kit includes the nucleotidase CD39L3 to quantitatively release inorganic phosphate from nucleotide diphosphate produced during the glycosyltransferase reaction. Because the amount of inorganic phosphate generated is proportional to the glycoconjugate product produced, included phosphate detection reagents can be used for accurate measurements of enzymatic activity. As an added benefit, the assay can be performed in multi-well plates and quantified by a plate reader, making it amenable to high-throughput screening.

Phosphatase-coupled Glycosyltransferase Reaction to Determine the Activity of Clostridium difficile Toxin B (TcdB).
A. The phosphatase releases inorganic phosphate from the nucleotide produced during the glycosyltransferase reaction. Because the phosphate released is proportional to the glycoconjugate product generated, phosphate levels can be used to examine enzyme activity.
B. Using UDP-Glc as a donor substrate, the Glycosyltransferase Activity Kit (#EA001) was used to measure the activity of the enzyme Recombinant TcdB (#6246-GT). The reaction was carried out in the presence of the phosphatase, Recombinant Human CD39L3, and free phosphate was detected using malachite .
